Try lubricating first. Push knobs should not rattle around excessively in the handle due to wear.
If that does not help go to Lin's excellent thread at Valvechatter for how to adjust. S type & Mk2 locks are not identical but can give some of the same issues. Click on images to blow up. There is a lug on the casting next to the adjuster screw that cracks & breaks off causing mayhem. Don't refit any cracked castings. Ignore remote control section unless interested in the mod. Stay on this page. Make sure that lock barrels are clean, lubricated & behaving. (see Fig 1 below)

Well if the barrel turns with the correct key then all the pins in the barrel are operating as designed. You are going to have to take off the interior handles, pop the door card fasteners & flap up the door card. The vinyl is glued to the top of the door.
Then remove the protective plastic & investigate the lock contact mechanism for a fault/damage, wear etc. and adjust the contactor clearance to contactor bolt to 1/32″
If this does not work you are going to have to remove the door handle and mechanism to find out where the issue is & repair.
Remember to reseal the door when you are finished to protect the card from water damage.
Good luck!

I had a similar problem on the front passengers door. The door was unlocked but the button when pushed would not open the door although it could be opened from the inside. It had occurred when I had mis set the door closing catch and the door needed a lot of pressure on the button to open it up. Before I had been able to realign the door catch, I pushed the button with force and it just went in and nothing happened.
I had to strip the door card and internals out to get to see how the lock worked and what had broken.
When you push the button in there is a pin/bracket on the back of the button which pushes a lever that operates the door catch. part of the button mech on mine due to my brute force and ignorance had snapped stopping the pin from coming in to contact with the lever.
There are three large cross head screws which release the catch but you also have to detach the internal door handle bar and locking bar before it comes out. This has to be removed so you can get to the small nuts which hold the door handle in place. Getting the door catch out of the door is then a struggle because the glass and window mech are in the way but it does come out. Once stripped it down this is what I found.
Unfortunately no way of fixing this unless you can weld aluminium. I was lucky I put out a call on the S Type forum and a friend of mine found a spare in a box in his garage and sent it to me. Replaced this part and it fixed the problem.

Yep. Similar to what I found on a couple of my locks (cracks starting). I had spares. That vertical lug next to where your crack started usually breaks off too. Seems a weak spot. A number of my spares had cracks starting at the base of that lug.
In fact if you look at Lin's picture above it looks as though some repair work might have been attempted at the base of the lug.
BTW Rob. Thanks for the nice picture that shows the issue clearly. Filed!
