Sounds like dead locking
Non U.S. spec cars have dead locking latches. The dead locking feature can be defeated by having the car programmed at the dealer with the IDS and disabling dead locking. If you are in the U.S. it is possible that dead locking has been programmed but without dead locking latches installed you should still be able to open the doors from the inside with just a pull of the handle. Were the handles stiff before the problem got to where you couldn't unlock door? May just need lube. Common problem.

XJ6 Door Lock Problem
We have a new to us XJ6 1997 that has bizarre door lock problems.
The locks on the front and rear passenger side work properly.
All of the manual push pull locking levers go fully up and down with the lock/unlock cycles.
The driver's side door can only be opened by using the outside handle. The inside handle fails regardless of the lock-unlock cycle.
The rear door on the driver's side cannot be opened using the inside or outside handles. The push pull lever moves properly. The child lock may be set on this door.

First thing to do is check your grounds. They are located at the bottom of the "A" pillers (front part of doorway, part of body not door). Bad grounds can make things real screwy. Clean them up as needed. Look for broken or almost broken wires. You may also have a problem with the GENERAL ELECTRONIC CONTROL MODULE. The CD JITS will show you where it is.

Door lock problems on 2007 x type
Hi I have a weird one too my front passenger door lock seems reluctant to allow my passenger to open it from inside unless the front seat is unoccupied! I have often been able to lean across & open it from my drivers seat either before or after I have opened my door. However its as if the passenger seat switch ( which I know is basically to operate the seat belt light & or notify the pass airbag whether there is a front passenger of a certain weight to deploy or not in the event of an accident!). Strange but if I get out of the car & open the passenger door from outside & re-shut it then it can be opened from inside.It does make me look good if I have a lady passenger as I looks as if I am being a gentleman getting out to open the door for them to get out but it is a pain in the a**e!
Any ideas folks as the jag dealer says its about £100 for a new lock & 3150 labour & I am not convinced that its the lock1 As It only seems to be un-openable when I have a front seat passenger !

I just started experiencing a weird problem over the weekend regarding my locks. One of the rear doors won't lock unless I push the lock lever in
by hand. It will unlock electronically but not lock. It seems that the lock lever
is somehow hanging up and the electronic motor in the door can't pull it
shut.
I looked at the lock lever and the door handle but don't see any interference. The door is rarely used so I don't think it got banged in any way.
Hate to have to pull the door panel off to see what's happening...

Weird door lock problem
I can think of only one thing... when you are in drive the locks won't unlock, a safety implementation I guess. So, when you put the car in Park, the locks can then be used to lock or unlock the doors. That said... It may be possible that your linkage or shifter safety switch is not being engaged, or disengaged, whichever the case may be. So try holding the gear shifter up towards the dashboard while you unlock the door. its only an outside chance, but you never know.

I too have a similar problem. But the rear doors are not affected at all. Only the two front doors. It used to be only the driver door, but the last month or so, the passenger door started to have the same issue At first, when I opened the doors with the fob you could close and open the passenger door with the doorhandle but now it wont to that either.
If I open and close the doors with the fob lots of time it suddenly works, but only for a minute then it is back to only the fob will open the doors again.

Its not the fob
I have tried with both my fobs, it's not the fob. It is when I try to open the doors from inside with the doorhandle. I just can't open them.
