Won't unlock with the key? Or won't unlock with the inside paddle? If just one or the other, the respective linkage may have just dropped off. You'd likely have the sensation of this...as the action would feel very loose.
If the door won't unlock with both the key and the inside paddle then the problem is probably with the door latch itself.....perhaps simply being gummed up and needing a good clean-and-lube treatment.
I dealt with this scenario a dozen or more years ago on a Series III I was repairing for a fellow. In that case the latch required replacement although the details of the actual failure escape my memory at the moment. It is possible to non-destructively remove the door card from inside the car. Not pleasant, but possible. Then, with manipulation of the levers and gizmos on the latch to unlock to door. I remember a pocket screwdriver being involved to lift a little lever.....but beyond that it's all foggy in my brain
