I don't think there is a way to test those. Inside it's GM 4 pin HEI module, so you can look and see if there is a test for those. They tend to either work or they don't.
Don't relocate the amp. It generates quite a bit of heat, and where it is is actually the coolest spot in the engine bay. The incomming air through the intake manifold acts as a very effective cooler. If you've ever driven a V12 in winter ( as I have) the engine block will be hot and the intake manifolds are very cold.

When they stop working, you'll know, as the engine will stop running and won't start until you replace the amp.
You only need to replace the HEI module. My experience has been that it's hit-or-miss on if a new module will work or not. I bought four, two of which worked and two which didn't. So I carry a working spare in the car. Being a GM part, they are fairly easy to find.

I just installed this NOS HEI Performance Ignition module for GM Cars 1974-1988 and Aftermarket Race Distributors. Supposed to provide a bit more dwell. We shall see. The good news is that she fires up nicely and revs smoothly with it. Some, as others have said above, simply don’t run the v12 at all.
