Yes if you have an ohmmeter you can check a fuse. If you don't have one, just swap it out with a new one. They're cheap enough.
It's possible your amp has failed. If the basics are confirmed to be good, then the next step is get diagnostic trouble codes (DTC's) from the car.
For that you will need a code reader that reads all codes. Cost to you is about $70 but worth it when you're on a forum. We use codes to help.
There are several code readers out there. Just don't go cheap as some will only read engine codes and you will need body codes for this.

While you are waiting.....locate the amp fuse as mentioned above and pull it. If its blown you can see it by holding it up to the light. Like Sean said, its a large 30amp fuse....so should be hard to miss.
Side note, On my old Range Rover, I had the sound cut out one day. All sources were dead. There were three different fuses that ran the infotainment system. I pulled them one by one, nothing. Then I pulled all three and let it do its thing. It worked.

@lear60man , well done. Thats a well known work around for the ML63, too. Essentially it boils down to resetting the module to factory defaults.
I don’t quite understand it though. That is, Powering the car off and on does not have the same effect as pulling the fuses.

I have had this happen on my 2010 XKR several times in my 8 year ownership as well as radio station presets don't appear on the screen, just blanks. Typically, either turning on and off the radio or restarting the car clears it up. Obviously, you've started your car many times since this started happening, so you must have a different issue.
