Welcome to the Forum. Now you have 2 questions with a whole realm of possibilities.
Quick Answer to your start question
To my knowledge, the XK8 will not start in Drive (D) and/or Reverse (R). I've only ever started the car from P and/or N. If you ever started from D and R - tell me more about it - as I would say that the gear stick linkage cable is wrongly adjusted!
Gearbox loss of Reverse -
First thing what Model Year (MY) car, spec ie convertible/coupe and is it a supercharged engine? Mileage would be nice and a service history esp. as you say it's had a new "box" which I presume you mean new gearbox. Plus you say fluid and filters are fine. How do you know all this? Did you do the work yourself? These automatic gearboxes are complicated, very sensitive and are super picky on fluid types and correct fill levels. What new gearbox bits went in - drive line - drums, clutches, valvebody, solenoids etc.
The X100 had 3 different gearboxes of which I'm familiar with the ZF5HP24 automatic transmission from the earlier MY cars. Losing Reverse may be the gear stick cable - as above, but this is a common problem in the ZF5HP24 where the typical reason for losing reverse is failure of the F drum clutch/piston. Other possibilities are defective pressure valves or even a broken valve body as fluid pressure transfer are in hydraulic channels in the valve body. These valves & channels can be cleaned and "tuned" by specialists.
More info is needed esp. in regard to the history of the car and your capabilities to DIY. Generally this transmission needs professional specialists.
